Charakterystyka pracy w języku polskim:

Celem pracy była funkcjonalna powierzchnia, która ułatwia odtworzenie naturalnej warstwy śródbłonka, która hamuje procesy koagulacji. W oparciu o konstrukcję i właściwości nisz komórkowych, które są naturalnym środowiskiem komórek macierzystych, podjęto próbę ich zrekonstruowania, stosując odpowiednią mikrostrukturę i topografię powierzchni. Zaprojektowano i wykonano ultra cienkie powłoki na bazie amorficznego węgla i węgla diamentopodobnego uzyskane metodami fizycznego osadzania z fazy gazowej (PVD). Skład powłok podyktowano wysokimi właściwościami hemokompatybilnych powłok węglowych i ich niską toksycznością. Topografaia powierzchni w formie fałd otrzymano przez kontrolowanie naprężeń własnych w powłoce, jak również mechanicznej deformacji podłoża przed nałożeniem powłok.

Materiały mogą być stosowane jako biomateriały przeznaczone do bezpośredniego kontaktu z krwią. Analiza interakcji materiał- krew w wykazała wysokie właściwości hemozgodne. Ocenę właściwoscici atrombogennych dokonano na podstawie badań dynamicznych. Wyniki wykazują bardzo dobre właściwości materiałów na bazie węgla amorficznego w kontakcie z krwią. Ostateczny wybór powłoki dokonano przez porównanie ilości apoptotycznych mikrocząstek uformowanych w funkcji zużycia płytek krwi, czyli ich niszczenia na skutek kontaktu z biomateriałem.

W badaniu poddano analizie nowe materiały do powlekania powierzchni implantów serca stosowanych w układach wspomagania krążenia, z uwzględnieniem ich projektowania, optymalizacji i testowania odpowiednich parametrów.

W pracy przedstawiono innowacyjną koncepcję modyfikacji powierzchni na bazie cienkich powłok o wysokich właściwościach hemozgodnych, modyfikowanych w kierunku struktur funkcjonujących symulujących nisze komórek macierzystych. Poprzez odtworzenie struktur fizjologicznych i funkcji tkanek określono rolę środowisk niszowych. Prawidłowo ukształtowane powierzchnie uzyskane metodami fizycznego osadzania z fazy gazowej wpływały na stopień różnicowania komórek i skuteczność tworzenia pojedynczych warstwy śródbłonka, która jest bezpośrednim inhibitorem procesu aktywacji i koagulacji krwi. Ultra cienkie powłoki wykazywały duży potencjał do kontrolowanego różnicowania komórek progenitorowych w kierunku śródbłonka.

The abstract of the thesis (in English):

The result of this work was a functional surface that facilitates reconstitution of the natural layer of normal endothelial tissue which inhibits the coagulation process. Based on the construction and properties of cellular niches, which are the natural environment of stem cells, attempts have been made to reconstruct them using appropriate microstructure and topography of the surface, based on surface interference lithography methods. Ultra-thin coatings based on amorphous carbon and diamond-like carbon deposited by physical vapor deposition (PVD) were designed and manufactured. The composition of the coatings was dictated by the high properties of hemocompatible carbon coatings and their low toxicity. Corrugated structures were obtained by controlling the amount of internal stresses in the coating as well as by mechanical deformation of the substrate prior to the application of the coatings.
Materials could be applied to items dedicated to direct contact with blood. The blood-material interaction analysis showed high haemostatic properties. The blood-material impact assessment was done on the basis of dynamic tests. The results show very good properties of amorphous carbon based materials in contact with blood. Final selection of the coating was made by comparing the amount of apoptotic microparticles formed as a function of platelet consumption.
In the study, new materials for coating the surface of cardiac implants used in forced blood circulatory systems, including design, optimization and testing of relevant parameters, were subjected to analysis. The subject matter of the work was dictated by the need for new materials for these devices that show improved interaction with the blood.
The work presents an innovative concept of surface modification based on thin pleated coatings with high hemomorphic properties, modified in the direction of structures that function as stem cell niches. By reconstructing physiological structures and tissue functions, the role of niche environments has been defined. Correctly shaped surfaces obtained by physical vapor deposition have affected the degree of cell differentiation and the efficiency of endothelial monolayer formation which is a direct inhibitor of the activation and coagulation processes. Ultra-thin folded coatings exhibited high potential for controlled differentiation of progenitor cells.
